What is gRNA

gRNA (guide RNA) is an RNA molecule used in genome-editing technologies, such as the CRISPR-Cas9 system. Along with the Cas enzyme, gRNA helps in recognizing and cleaving specific DNA sequences. Due to its customizability, efficiency, and versatility, gRNA is an essential component of genome-editing technologies.

Although the length of the most commonly used gRNA used
in CRISPR-Cas9 system is 100 mer,
recent advanced genome-editing technologies use even longer gRNAs.

Purity is a Critical Factor

When it comes to gRNA for therapeutic applications, purity is a critical factor.

  • For therapeutic applications, regulatory agencies will look for the use of reagents at the best available purity, regardless of in-vivo or ex-vivo use.

    Former Executive, Biotech

  • Regulators now require at least 80% purity and a well-designed analytical control strategy.

    VP, Quality, Biotech

  • “High purity is important.” “Using 1.5 times the required quantity of a 60% pure gRNA does not provide the same activity as a 90% pure gRNA.”

    Professor at an academic institution

  • There are multiple advantages in using high purity gRNA—easier to work with, easier to characterize, and lower probability of off-target effects.

PMM chemistry significantly

In case of 100 mer synthesis, TBDMS chemistry produces about four times as many 2’, 3’-switched isomers as PMM chemistry.

TBDMS group is more labile than PMM group under acidic condition. Thus, TBDMS chemistry leads to more 2’, 3’-switched isomers as described in figure 1*.

FAQs

  • Q.What is the maximum length of gRNA that can be produced?
A.200 mer is the longest we have synthesized. From that data, we are confident of synthesizing longer oligonucleotides. If there is a requirement, please contact us.
  • Q.How can we verify the quality of your gRNA?
A.We can provide a small-scale sample of a specific sequence free of charge.
  • Q.How do you prove the high purity of your gRNA?
A.Please refer to the following document for information on our strong analytical methods. (Link to download the document)
  • Q.What capacity of gRNA production can you handle?
A.We can handle both large-scale GMP production and small-scale production. The quantity of the product depends on the sequence; however, for 100 mer gRNA, we have produced 15g at half scale and 30g at full scale. If you need a larger quantity, we can combine after the purification process. For small-scale production, we can share the optimal production schedule and cost, including leveraging our alliances. Please contact us.
  • Q.Do you perform sequence tests?
A.Sequence tests can also be conducted in-house. We would be happy to discuss this further with you.
  • Q.Where is the site for non-GMP and GMP manufacturing?
A.All manufacturing processes, both non-GMP and GMP, occur in Japan.
  • Q.Can you assist with customs clearance?
A.We have consistently encountered no issues with customs. Sumitomo Chemical will provide full support regarding all necessary documentation for customs.
  • Q.What is the FDA inspection history of Oita Works?
A.Although our gRNA facility is yet to be inspected by the FDA, the same GMP system as the small molecule facility at Oita Works is applied. Small molecule facility at Oita works has successfully passed numerous FDA inspections.
